چکیده

UNLABELLED PREMISE OF THE STUDY Firmiana consists of 12-16 species, many of which are narrow endemics. Expressed sequence tag (EST)-simple sequence repeat (SSR) markers were developed and characterized for size polymorphism in four Firmiana species. • METHODS AND RESULTS A total of 102 EST-SSR primer pairs were designed based on the transcriptome sequences of F. danxiaensis; these were then characterized in four Firmiana species-F. danxiaensis, F. kwangsiensis, F. hainanensis, and F. simplex. In these four species, 17 primer pairs were successfully amplified, and 14 were polymorphic in at least one species. The number of alleles ranged from one to 13, and the observed and expected heterozygosities ranged from 0 to 1 and 0 to 0.925, respectively. The lowest level of polymorphism was observed in F. danxiaensis. • CONCLUSIONS These polymorphic EST-SSR markers are valuable for conservation genetics studies in the endangered Firmiana species.

De Novo Transcriptome Assembly in Firmiana danxiaensis, a Tree Species Endemic to the Danxia Landform

Many Firmiana species are locally endemic, providing an interesting system for studying adaptation and speciation. Among these species, F. danxiaensis is a tree species endemic to Mount Danxia in Guangdong, China, which is an area known for presenting the Danxia landform. How F. danxiaensis could have adapted to the stressful environment of rocky cliffs covered with barren soils in the Danxia l...

Transcriptome analysis of the freshwater pearl mussel, Hyriopsis cumingii (Lea) using illumina paired-end sequencing to identify genes and markers

The transcriptome of triangle sail mussel Hyriopsis cumingii (Lea) using Illumina paired-end sequencing technology was conducted and analyzed. Equal quantities of total RNA isolated from six tissues, including gonad, hepatopancreas, foot, mantel, gill and adductor muscle, were pooled to construct a cDNA library. A total of 58.09 million clean reads with 98.48 % Q20 bases were generated. Cluster...

Application of Random Amplified Microsatellite Polymorphism (RAMP) in Prunus Characterization and Mapping

Random amplified microsatellite polymorphism (RAMP) is a PCR-based marker which uses a combination of two classes of markers: Simple sequence repeat (SSR) and Random amplified DNA polymorphism (RAPD) markers. RAMP has been demonstrated to be a potentially valuable molecular marker for the study of genetic relationships in cultivated plant species. The objective of this study was to optimize the...

Isolation and characterization of 13 new microsatellite markers in the triangle mussel (Hyriopsis cumingii)

Microsatellite enriched library of Hyriopsis cumingii was constructed according to the strong affinity between biotin and streptavidin. One hundred clones with foreign inserts were sequenced and 65 clones were screened for usable microsatellites, of which 36 were deemed unique, of sufficient length (more than 8 repeats) and possessed adequate flanking regions for primer designment. Among 36 pri...
